注意事項(xiàng) | 1.PC-12(Undifferentiated)細(xì)胞正常生長狀態(tài)為懸浮聚團(tuán),若需要用NGF誘導(dǎo)時(shí),需要使用多聚-L-賴氨酸溶液(貨號 PB180523)包被培養(yǎng)瓶后促使細(xì)胞貼壁。2.Undifferentiated、Low differentiation和High differentiation的區(qū)別:Undifferentiated的PC-12從形態(tài)上看是圓形的,聚團(tuán)生長的漂浮細(xì)胞;Low differentiation的成多角形,有較短的突起;High differentiation的細(xì)胞有多個(gè)突起,突起數(shù)目不等,突觸較長,類似神經(jīng)元軸突。3.Low differentiation和High differentiation是在Undifferentiated的PC-12基礎(chǔ)上誘導(dǎo)產(chǎn)生的,Low differentiation和High differentiation指的與神經(jīng)細(xì)胞表型的相似程度,High differentiation更接近神經(jīng)細(xì)胞表型。 |
背景描述 | PC-12(Undifferentiated)細(xì)胞是來自能移植的雄性大鼠腎上腺嗜鉻細(xì)胞瘤。PC-12(Undifferentiated)細(xì)胞表達(dá)神經(jīng)生長因子(NGF)受體,NGF可誘導(dǎo)產(chǎn)生神經(jīng)表型。PC-12(Undifferentiated)細(xì)胞不合成腎上腺素。 |
