注意事項 | 22RV1細胞復(fù)蘇時需要離心去除DMSO。凍存細胞復(fù)蘇最初階段,貼壁量少,成簇松散貼壁,但是最終細胞會變平并且擴散成很好的單層,這個過程需要4-5天時間。細胞生長緩慢,細胞只能生長到90%的匯合度。細胞凍存后復(fù)蘇成活率不高,推薦凍存液配比為90%FBS+10% DMSO,使用優(yōu)質(zhì)血清。 |
背景描述 | 22RV1是來自異種移植(在閹割引起前列腺癌衰退又在其父親的雄性激素信賴型CWR22嫁接后復(fù)發(fā)的小鼠中連續(xù)傳代)的人前列腺癌上皮細胞系;22RV1細胞系表達前列腺特異抗原。二羥基睪丸脂酮輕微刺激22RV1細胞生長,經(jīng)Western Blot檢測溶解產(chǎn)物與抗雄性激素受體抗體起免疫反應(yīng);EGF刺激22RV1細胞生長,但TGFβ-1不能抑制細胞生長;22RV1可在裸鼠中成瘤。 |
