  5. 細胞簡介:

    人臍靜脈內(nèi)皮細胞分離自臍帶組織;它是臍靜脈的重要結構組成細胞之一,在機體的正常生理過程中發(fā)揮著重要作用。臍帶是哺乳類的連接胎兒和胎盤的管狀結構,臍帶中通過尿膜的血管即臍動脈和臍靜脈,卵黃囊的血管即臍腸系膜動脈及臍腸系膜靜脈。在子宮中,子宮動脈在胎盤的母體部分出的毛細血管,與胎盤的子體部胎兒毛細血管靠近,在此處母體和胎兒的血液間進行CO2和O2,代謝產(chǎn)物即代謝廢物和營養(yǎng)物質(zhì)的交換。臍動脈將胎兒產(chǎn)生的廢物運送至胎盤,臍靜脈將O2和營養(yǎng)物質(zhì)從胎盤運送給胎兒。

  6. 方法簡介:

    普諾賽實驗室分離的人臍靜脈內(nèi)皮細胞采用胰蛋白酶-膠原酶聯(lián)合消化法結合差速貼壁法、并通過內(nèi)皮細胞專用培養(yǎng)基培養(yǎng)篩選制備而來,細胞總量約為5×10?cells/瓶。

  7. 質(zhì)量檢測:

    普諾賽實驗室分離的人臍靜脈內(nèi)皮細胞經(jīng)CD31免疫熒光鑒定,純度可達90%以上,且不含有HIV-1、HBV、HCV、支原體、細菌、酵母和真菌等。

  8. 培養(yǎng)信息:

    包被條件PLL(0.1mg/ml),明膠(0.1%)
    培養(yǎng)基含F(xiàn)BS、生長添加劑、Penicillin、Streptomycin等

    換液頻率每2-3天換液一次
    生長特性貼壁
    細胞形態(tài)內(nèi)皮細胞樣
    傳代特性可傳3-5代左右;3代以內(nèi)狀態(tài)最佳
